Yes, I checked out the Ebay links and they both definitely are preprinted.  It is interesting that he did 2 variations.

Jennifer, I did not intend to sound rude. And, I can understand the confusion. I did a search last night on John Wayne signed business cards and found a number of them listed around the Internet being offered as authentic with very high prices. Each had a story that Mr. Wayne took the card out and signed them in-person. But, from everything I can tell, this did not happen, at least not on any frequent basis. I cannot find one example that looks to be signed by John Wayne in-person. Sorry.

Pre-print signature. You can clearly see air spots.
